pg 413 of PPH (Interaction with Other Drugs): There is generally no need to cease taking other drugs in the preceding days. Nevertheless, it is common practice for those planning their death to cease all but the essential medication in the week before their planned exit.
PPH also says that prolonged use of anti-psychotic meds or chronic, heavy alcohol use is associated with prolonged coma stage. It's the last paragraph of pg 410 (pdf). My personal take was that a second bottle is used as a layer of certainty for successful outcome.
The anti psychotics will slow down the absorption of N. The N would still work, but you risk going into an extended coma. The PPH recommends that you stop taking the anti psychotics for at least 2 weeks before taking N if you can.
